Not only do I have a family of six myself, I also grew up in a family of six, so I am well aware of the challenges of finding accommodations for larger families! Most standard Walt Disney World Hotel rooms sleep 4 or 5 Guests, plus one child under the age of 3 in a crib.
Disney's Art of Animation and
Disney's All-Star Music Resorts offer Family Suites that do accommodate 6 Guests. Family Suites at both resorts have two bathrooms, as well as a kitchenette with a small refrigerator, microwave, and coffeemaker.
Wilderness Cabins at Disney's Fort Wilderness Resort can also sleep six, have one full bathroom and also a full kitchen with cooking appliances and utensils.
Deluxe Resort Hotels, such as Disney's BoardWalk Inn, Disney's Wilderness Lodge and Disney's Yacht and Beach Club Resorts offer some Club Level Deluxe Rooms that sleep six with two Queen beds and a Queen-size sleeper sofa, as well as some larger suites. There are also 2-Bedroom Deluxe Villa accommodations that sleep up to 9 Guests. My friend and fellow panelist, (and mom of five!)

I have found that often the most economical option for my family of 6 is to reserve two standard rooms instead of a suite. This gives us plenty of space and beds, and two bathrooms, which is super helpful with a larger family. It is possible to reserve two rooms at any Disney Resort Hotel, and request connecting rooms. While requests cannot be guaranteed, in my experience Cast Members will do their best to honor your request. I recommend

Disney Resort Hotel that meets your needs.
